dc.description.resumoOs compostos presentes na erva-mate são reconhecidos pelos benefícios à saúde, destacando-se a atividade quimio-preventiva, a inibição do estresse oxidativo, a propulsão intestinal e o efeito vasodilatador e colerético. Dentre os compostos bioativos presentes na erva-mate, destacam-se os alcaloides metilxantínicos, especialmente a cafeína e teobromina, reconhecidos por suas propriedades estimulantes do sistema nervoso central e os compostos fenólicos, com ação antioxidante. Assim, avaliar se a tecnologia de extração supercrítica é efetiva na obtenção de extratos de folhas, caule e flor de erva-mate, bem como as características dos extratos obtidos foram os principais objetivos deste trabalho. Para isso, as condições de pressão e temperaturas de extração foram otimizadas quanto ao rendimento global (R), rendimento na taxa de extração constante (RCER) e tempo de extração na taxa constante (tCER) para as folhas de erva-mate. A pressão de operação do sistema ES-CO2 influenciou positivamente na obtenção de extratos de folhas de erva-mate. A 32 MPa e 58 °C foi possível obter 8,78 % de extrato em relação a massa de matéria-prima. Quando aplicada esta condição a caules e flores, obteve-se rendimentos de extração de 1,37 % e 2,24%, respectivamente. As cinéticas de extração foram avaliadas através do modelo Spline, que considera três etapas de extração. O modelo ajustou-se adequadamente aos dados experimentais, apresentando coeficientes de determinação (R2) superiores a 0,99. Os tempos da etapa de taxa constante de extração foram determinados entre 23,7 e 60,6 min, sendo esta epara responsável pela obtenção de cerca 40 % do total de extrato. A quantificação e identificação dos extratos foram avaliadas por cromatografia líquida de alta eficiência (CLAE) e FTIR. Os principais compostos identificados foram as metilxantinas cafeína e teobromina. Os extratos de flores, caules e flores apresentaram 35,5 e 5,5 mg/g, 63,9 e 9,6 mg/g e 43,1 e 48 mg/g de cafeína e teobromina, respectivamente. Entretanto, os extratos apresentaram baixa atividade antioxidante. Em condições otimizadas a ES-CO2 é uma técnica viável para a obtenção de extratos de erva-mate especialmente para a extração de compostos de média polaridade, como a cafeína e teobromina, com potencial aplicação em alimentos, contribuindo para a valoração da cadeia produtiva deste produto..por
dc.description.abstractThe compounds present in the yerba mate are recognized for their health benefits, with emphasis on chemo-preventive activity, inhibition of oxidative stress, intestinal propulsion and the vasodilator and choleretic effect. Among the bioactive compounds present in yerba mate, methylxanthine alkaloids, especially caffeine and theobromine, are well known for their central nervous system stimulating properties and phenolic compounds with antioxidant action. Thus, to evaluate if the technology of supercritical extraction is effective in obtaining extracts of leaves, stem and flower of mate, as well as the characteristics of the extracts obtained were the main objectives of this work. For this, the pressure conditions and extraction temperatures were optimized for the overall yield (R∞), yield at constant extraction rate (RCER) and time at constant rate extraction (tCER) for mate leaves. The operating pressure of the ES-CO2 system positively influenced the extraction of extracts of mate leaves. At 32 MPa and 58 ° C it was possible to obtain 8.78% of extract in relation to the mass of raw material. When this condition was applied to stems and flowers, extraction yields of 1.37% and 2.24%, respectively, were obtained. The extraction kinetics were evaluated through the Spline model, which considers three stages of extraction.
